One may expect that this technology in the future will ensure the creation of an operational and highly effective procedure for forensic DNA profiling.</p> <p><bold><italic>MATERIAL AND METHODS:</italic></bold> The methodical basis of the work is the genotyping of polymorphic STR-loci of chromosomal DNA in biological material using high-tech fully automated (full cycle) analytical platform RapidHIT: namely, RapidHIT 200 (Integenx, USA) and RapidHIT-ID (Life Technologies/Thermo Fisher, IntegenX (USA) instruments.</p> <p><bold><italic>RESULTS: </italic></bold>Buccal epithelium and blood samples, while being the most common in a forensic DNA testing, have been examined as a validation test objects. Then experiments were carried out with real expert material such as cigarette butts, hair, nails, bones, chewing gum; swabs from different surfaces were examined as test samples as well.</p> <p><bold><italic>CONCLUSIONS: </italic></bold>Based on the data obtained, it can be stated that the Rapid DNA technology as it is implemented in the full-cycle genetic analyzers IntegenX RapidHIT 200 (Integenx, USA) and RapidHIT ID (Thermo Fisher, IntegenX, USA), according to the criteria studied, in general supports the idea of to be introduced into the practice of forensic DNA analysis.</p></abstract><trans-abstract xml:lang="ru"><p><bold><italic>Обоснование.</italic></bold> Технология «быстрой ДНК» в перспективе станет оперативной и высокоэффективной процедурой судебно-экспертного молекулярно-генетического исследования, направленного на получение генетической информации о принадлежности биологического материала конкретному лицу.</p> <p><bold><italic>Цель исследования. </italic></bold>Работа представляет собой часть комплексных исследований, проводимых ФГБУ РЦСМЭ Минздрава России, конечной целью которых является развитие новых высокоэффективных технологий молекулярно-генетической индивидуализации и внедрение их в сферу деятельности отечественной судебно-медицинской экспертизы.</p> <p><bold><italic>Материал и методы. </italic></bold>Методической основой работы является генотипирование полиморфных STR-локусов хромосомной ДНК (анализ полиморфизма длины амплифицированных фрагментов хромосомной ДНК) в объектах биологической природы с помощью высокотехнологичных автоматизированных генетических анализаторов полного цикла. В настоящей работе использованы станции IntegenX RapidHIT 200 (IntegenX, США) и RapidHIT ID (Thermo Fisher-IntegenX, США).</p> <p><bold><italic>Результаты. </italic></bold>В качестве валидационных тест-объектов, которые были проанализированы с помощью автоматизированных генетических анализаторов полного цикла, исследованы образцы буккального эпителия и крови как наиболее распространённые в судебно-медицинской практике. Далее были выполнены эксперименты с реальными объектами экспертного исследования: в качестве тест-образцов были исследованы сигаретные окурки, волосы, ногти, кости, жевательная резинка, смывы биоматериала с различных поверхностей.
